Amplified rDNA restriction analysis of clones from the libraries indicate that the rDNA richness and evenness was high: A few clones also grouped with the Betaproteobacteria, Nitrospirae, Spirochaetales, Chlamydiae, Firmicutes, and candidate division OPl 1. The abundance of sequences affiliated with Gammaproteobacteria was higher in libraries from shallow sediments in the Thermaikos Gulf 30 m and the Cretan Sea m compared to the deeper South Ionian station m.
Most sequences in the four sediment libraries clustered with uncultured 16S rDNA phylotypes from marine habitats, and many of the closest matches were clones from hydrocarbon seeps, benzene-mineralizing consortia, sulfate reducers, sulk oxidizers, and ammonia oxidizers. LIBSHUFF statistics of 16S rDNA gene sequences from the four libraries revealed major differences, indicating either a very high richness in the sediment bacterial communities or considerable variability in bacterial community composition among regions, or both.
Mobile phone applications and the utilization of library services in Mobile phone applications and the utilization of library services in the university of Calabar library , Calabar, Nigeria. Global Journal of Educational Research Diazotrophs are key players of the globally important biogeochemical nitrogen cycle, having a significant role in maintaining ecosystem sustainability. Saline soils are pristine and unexplored habitats representing intriguing ecosystems expected to harbour potential diazotrophs capable of adapting in extreme conditions, and these implicated organisms are largely obscure.
Differential occurrence of diazotrophs was studied by the nifH gene-targeted clone library approach. Four nifH gene clone libraries were constructed from different soil niches, that is saline soils low and high salinity; EC 3. Additionally, the abundance of diazotrophic community members was assessed using quantitative PCR.

Analysis of microbiota associated with peri-implantitis using 16S rRNA gene clone library. Full Text Available Background: Peri-implantitis PI is an inflammatory disease which leads to the destruction of soft and hard tissues around osseointegrated implants.
The subgingival microbiota appears to be responsible for peri-implant lesions and although the complexity of the microbiota has been reported in PI, the microbiota responsible for PI has not been identified. The purpose of this study was to identify the microbiota in subjects who have PI, clinically healthy implants, and periodontitis-affected teeth using 16S rRNA gene clone library analysis to clarify the microbial differences.
Three subjects participated in this study. The conditions around the teeth and implants were evaluated based on clinical and radiographic examinations and diseased implants, clinically healthy implants, and periodontally diseased teeth were selected. Subgingival plaque samples were taken from the deepest pockets using sterile paper points.
Prevalence and identity of bacteria was analyzed using a 16S rRNA gene clone library technique. A total of different species were identified from clones sequenced. The numbers of bacterial species identified at the sites of PI, periodontitis, and periodontally healthy implants were 77, 57, and 12, respectively. Microbiota in PI mainly included Gram-negative species and the composition was more diverse when compared to that of the healthy implant and periodontitis.
The phyla Chloroflexi, Tenericutes, and Synergistetes were only detected at PI sites, as were Parvimonas micra, Peptostreptococcus stomatis, Pseudoramibacter alactolyticus, and Solobacterium moorei. Low levels of periodontopathic bacteria, such as Porphyromonas gingivalis and Aggregatibacter actinomycetemcomitans, were seen in peri-implant lesions. The biofilm in PI showed a more complex microbiota when compared to periodontitis and.

High-dimensional quantum cloning and applications to quantum hacking. Attempts at cloning a quantum system result in the introduction of imperfections in the state of the copies.
This is a consequence of the no- cloning theorem, which is a fundamental law of quantum physics and the backbone of security for quantum communications. Although perfect copies are prohibited, a quantum state may be copied with maximal accuracy via various optimal cloning schemes. Optimal quantum cloning , which lies at the border of the physical limit imposed by the no-signaling theorem and the Heisenberg uncertainty principle, has been experimentally realized for low-dimensional photonic states.
However, an increase in the dimensionality of quantum systems is greatly beneficial to quantum computation and communication protocols. Nonetheless, no experimental demonstration of optimal cloning machines has hitherto been shown for high-dimensional quantum systems. We perform optimal cloning of high-dimensional photonic states by means of the symmetrization method.
We show the universality of our technique by conducting cloning of numerous arbitrary input states and fully characterize our cloning machine by performing quantum state tomography on cloned photons. In addition, a cloning attack on a Bennett and Brassard BB84 quantum key distribution protocol is experimentally demonstrated to reveal the robustness of high-dimensional states in quantum cryptography.
Molecular cloning , expression, and characterization of four novel thermo-alkaliphilic enzymes retrieved from a metagenomic library. Enzyme discovery is a promising approach to aid in the deconstruction of recalcitrant plant biomass in an industrial process. Novel enzymes can be readily discovered by applying metagenomics on whole microbiomes. Our goal was to select, examine, and characterize eight novel glycoside hydrolases that were previously detected in metagenomic libraries , to serve biotechnological applications with high performance. Here, eight glycosyl hydrolase family candidate genes were selected from metagenomes of wheat straw-degrading microbial consortia using molecular cloning and subsequent gene expression studies in Escherichia coli.
These proteins, denoted as proteins 1, 2, 5 and 6, were his-tag purified and their nature and activities further characterized using molecular and activity screens with the p NP-labeled substrates. Within a digital library , collections may range from an ad hoc set of objects that serve a temporary purpose to established library collections intended to persist through time. The objects in these collections vary widely, from library and data center holdings to pointers to real-world objects, such as geographic places, and the various metadata schemas that describe them.
The key to integrated use of such a variety of collections in a digital library is collection metadata that represents the inherent and contextual characteristics of a collection. An analysis of bacterial flora of the first voided urine specimens of patients with male urethritis using the 16S ribosomal RNA gene-based clone library method. To analyse the bacterial flora of urine from patients with male urethritis using the clone library method.
Urine specimens from patients with urethritis were used. The bacterial flora was analysed according to the 16S ribosomal RNA gene-based clone library method. Among 58 urine specimens, 38 were successfully analysed using the clone library method. From the specimens, clones were evaluated and 95 bacterial phylotypes were detected. In addition, Haemophilus influenzae and Neisseria meningitidis were also detected in 8 and 1 specimens, respectively.

After stable transfection of cDNA-expression- libraries G resistant clones were selected for enhanced radioresistance by a fractionated X-ray selection. One surviving transfectant clone clone exhibited enhanced radiation resistance in dose-response experiments and further X-ray selections.
Cell cycle analysis revealed complementation of untreated and irradiated cells in cell cycle progression. The rate of DNA synthesis, however, is not diminished after irradiation but shows the reverse effect.
The cDNA-sequence could be localized to human chromosome After stable transfection into AT-cells, one cosmid- clone was able to confer enhanced radiation resistance both in X-ray selections and dose-response experiments. The results indicate that the cloned cDNA-fragment is based on an unknown gene from human chromosome 11 which partially complements the radiosensitivity and the defective cell cycle progression in AT5BIVA cells.
Production of application libraries for WIMS. The WIMS codes have been developed over a period of 30 years to include sophisticated modelling of many thermal reactor lattices. A new cross-section library is to be released with WIMS6. Quality Assurance into the XMAS library is achieved in three levels; through general procedures during assembly, by validation studies and by verification. The main general procedure involves forming overall physics quantities Thermal average cross section, resonance integrals and fission spectrum average values from the point evaluations and comparing them with similar quantities from the XMAS library.
Examples of validation are studies optimizing the background cross-sections at which resonance integrals are tabulated and comparing reaction rates from homogeneous calculations with those formed in Monte Carlo and deterministic codes which represent energy variation of cross-section on a hyperfine energy mesh. Verification involves performing benchmark calculations and comparing results with experiment and calculations with the Apollo code. Construction characterization and application of Flavivirus infectious clones. The topics in this thesis revolve around a group of plus-strand RNA viruses that belong to the Flavivirus genus, a general introduction of which is presented in Chapter 1.
The experimental chapters in this thesis mainly focus on the construction and characterization of flavivirus infectious clones. Special libraries in Slovenia represent an important part of information infrastructure. Our government is undoubtedly aware of the deficiency of normative regulation in these areas; however, passing of certain acts is often delayed because there is no direct damage to the state itself if they Despite of ever increasing scope and volume of online information content and the constant need for easy accessibility to these contents, experts in various subject matters always face time constraints when searching for information or looking up their intended resource.
Phylogenetic characterization of a biogas plant microbial community integrating clone library 16S-rDNA sequences and metagenome sequence data obtained by pyrosequencing. The phylogenetic structure of the microbial community residing in a fermentation sample from a production-scale biogas plant fed with maize silage, green rye and liquid manure was analysed by an integrated approach using clone library sequences and metagenome sequence data obtained by pyrosequencing.
Sequencing of clones from a bacterial and an archaeal 16S-rDNA amplicon library revealed that the obtained nucleotide sequences are similar but not identical to 16S-rDNA database sequences derived from different anaerobic environments including digestors and bioreactors. Most of the bacterial 16S-rDNA sequences could be assigned to the phylum Firmicutes with the most abundant class Clostridia and to the class Bacteroidetes, whereas most archaeal 16S-rDNA sequences cluster close to the methanogen Methanoculleus bourgensis.
Further sequences of the archaeal library most probably represent so far non-characterised species within the genus Methanoculleus. A similar result derived from phylogenetic analysis of mcrA clone sequences. The mcrA gene product encodes the alpha-subunit of methyl-coenzyme-M reductase involved in the final step of methanogenesis.
Moreover, a large fraction of 16S-rDNA metagenome reads could not be assigned to lower taxonomic ranks, demonstrating that numerous microorganisms in the analysed fermentation sample of the biogas plant are still unclassified or unknown. Pulse crops are considered minor on a global scale despite their nutritional value for human consumption. Therefore, they are relatively less extensively studied in comparison with the major crops.
The need to improve pulse crop production and quality will increase with the increasing global demand for food security and people's awareness of nutritious food. The improvement of pulse crops will require fully utilizing all their genetic resources. Bacterial artificial chromosome BAC libraries of pulse crops are essential genomic resources that have the potential to accelerate gene discovery and enhance molecular breeding in these crops. Here, we review the availability, characteristics, applications , and potential applications of the BAC libraries of pulse crops.
The first insight into the salvia lamiaceae genome via bac library construction and high-throughput sequencing of target bac clones. Salvia is a representative genus of Lamiaceae, a eudicot family with significant species diversity and population adaptibility. One of the key goals of Salvia genomics research is to identify genes of adaptive significance. This information may help to improve the conservation of adaptive genetic variation and the management of medicinal plants to increase their health and productivity.
Large-insert genomic libraries are a fundamental tool for achieving this purpose. We report herein the construction, characterization and screening of a gridded BAC library for Salvia officinalis sage. Seventeen positive clones average insert size Kb containing five terpene synthase TPS genes were screened out by PCR and 12 of them were subject to Illumina HiSeq sequencing, which yielded 28,, bp raw reads 2. Scaffolds containing sabinene synthase Sab , a Sab homolog, TPS3 kaurene synthase-like 2 , copalyl diphosphate synthase 2 and one cytochrome P gene were retrieved via de novo assembly and annotation, which also have flanking noncoding sequences, including predicted promoters and repeat sequences.
Among 2, repeat sequences, there are amplifiable microsatellites. This BAC library provides a new resource for Lamiaceae genomic studies, including microsatellite marker development, physical mapping, comparative genomics and genome sequencing. Characterization of positive clones provided insights into the structure of the Salvia genome. These sequences will be used in the assembly of a future genome sequence for S. PDBlib forms the kernel of a larger software framework being developed for assiting in knowledge discovery from macromolecular structure data.
The software design strategy used by PDBlib, how the library may be used and several prototype applications that use the library are summarized. However, the memory resident representation of the macromolecule is independent of the PDB entry and can be obtained from other back-end data sources, for example, existing relational databases and our own object oriented database OOPDB built on top of the commercial object oriented database, ObjectStore.
At the front-end are several prototype applications that use the library: Macromolecular Query Language MMQL is based on a separate class library MMQLlib for building complex queries pertaining to macromolecular structure; PDBtool is an interactive structure verification tool; and PDBview, is a structure rendering tool used either as a standalone tool or as part of another application. Each of these software components are described. All software is available via anonymous ftp from cuhhca. Bovine viral diarrhea virus: Escherichia coli was transformed with the recombinant plasmids and a library of approximately BVDV specific cDNA clones varying in length from 0.
Probes did not hybridize with uninfected cell culture or cellular RNA. Hybridization probes were at least as sensitive as infectivity assays in detecting homologous virus. Complementation of the UV-sensitive phenotype of a xeroderma pigmentosum human cell line by transfection with a cDNA clone library. In previous work, a xeroderma pigmentosum cell line belonging to complementation group C was established by transformation with origin-defective simian virus We now report the complementation of the UV sensitivity of this cell line by gene transfer.
A human cDNA clone library constructed in a mammalian expression vector, and itself incorporated in a lambda phage vector, was introduced into the cells as a calcium phosphate precipitate. Following selection to G resistance, provided by the neo gene of the vector, transformants were selected for UV resistance. Twenty-one cell clones were obtained with UV-resistance levels typical of normal human fibroblasts. All transformants contained vector DNA sequences in their nuclei. Upon further propagation in the absence of selection for G resistance, about half of the primary transformants remained UV-resistant.
Secondary transformants were generated by transfection with a partial digest of total chromosomal DNA from one of these stable transformants. This resulted in 15 Gresistant clones , 2 of which exhibited a UV-resistant phenotype. The other primary clones lost UV resistance rapidly when subcultured in the absence of G Importantly, several retained UV resistance under G selection pressure. The acquisition of UV resistance by secondary transformants derived by transfection of DNA from a stable primary transformant, and the linkage between G and UV resistances in the unstable primary transformants, strongly suggests that the transformants acquired UV resistance through DNA-mediated gene transfer and not by reversion.
Among numerous applications , the polymerase chain reaction PCR 1,2 provides a convenient means to clone 5' ends of rare mRNAs and to generate cDNA libraries from tissue available in amounts too low to be processed by conventional methods.

Basically, the amplification of cDNAs by the PCR requires the availability of the sequences of two stretches of the molecule to be amplified. A sequence can easily be imposed at the 5' end of the first-strand cDNAs corresponding to the 3' end of the mRNAs by priming the reverse transcription with a specific primer for cloning the 5' end of rare messenger or with an oligonucleotide tailored with a poly dT stretch for cDNA library construction , taking advantage of the poly A sequence that is located at the 3' end of mRNAs.
We 3 and others have described strategies based on the addition of a homopolymeric dG 4,5 or dA 6,7 tail using terminal deoxyribonucleotide transferase TdT "anchor-PCR" [4]. However, this strategy has important limitations. The TdT reaction is difficult to control and has a low efficiency unpublished observations. To circumvent these drawbacks, we have used two approaches.
First, we devised a strategy based on a cRNA enrichment procedure, which has been useful to eliminate nonspecific-PCR products and to allow detection and cloning of cDNAs of low abundance 3. More recently, to avoid the nonspecific amplification resulting from the annealing of the homopolymeric tail oligonucleotide, we have developed a novel anchoring strategy that is based on the ligation of an oligonucleotide to the 35' end of ss. Nondestructive-assay applications may exploit this phenomenon passively, for example, in the presence of spontaneous-fission neutrons, or actively where an external neutron source is used as a probe.
In this work, we describe how new thermal neutron-capture data are being used to improve information in the neutron-data libraries for isotopes relevant to nonproliferation applications. We address this problem by providing new experimentally-deduced partial and total neutron-capture reaction cross sections and then evaluate these data by comparison with statistical-model calculations.

The purpose is to provide an overview Full Text Available The recent introduction of the WebGL API for leveraging the power of 3D graphics accelerators within Web browsers opens the possibility to develop advanced graphics applications without the need for an ad-hoc plug-in. There are several contexts in which this new technology can be exploited to enhance user experience and data fruition, like e-commerce applications , games and, in particular, Cultural Heritage.
In fact, it is now possible to use the Web platform to present a virtual reconstruction hypothesis of ancient pasts, to show detailed 3D models of artefacts of interests to a wide public, and to create virtual museums. SpiderGL provides data structures and algorithms to ease the use of WebGL, to define and manipulate shapes, to import 3D models in various formats, and to handle asynchronous data loading. We show the potential of this novel library with a number of demo applications and give details about its future uses in the context of Cultural Heritage applications. The neutron capture reaction is useful in identifying and analyzing the gamma-ray spectrum from an unknown assembly as it gives unambiguous information on its composition.
This can be done passively or actively where an external neutron source is used to probe an unknown assembly. There are known capture gamma-ray data gaps in the ENDF libraries used by transport codes for various nuclear applications.
For medium to heavy nuclei the quasi continuum contribution to the gamma cascades is not experimentally resolved. This code also provides a consistency check of the level scheme nuclear structure evaluation. The calculated continuum is of sufficient accuracy to include in the ENDF libraries.
This analysis also determines new total thermal capture cross sections and provides an improved RIPL database. For higher energy neutron capture there is less experimental data available making benchmarking of the modeling codes more difficult. We are investigating the capture spectra from higher energy neutrons experimentally using surrogate reactions and modeling this with Hauser-Feshbach codes. This can be used to simulate spectra from neutron capture at incident neutron energies up to 20 MeV to improve the gamma-ray spectrum in neutron data libraries used for transport modeling of unknown assemblies.
They are investigating the capture spectra from higher energy neutrons experimentally using surrogate reactions and modeling this with Hauser-Feshbach codes. Today's scientific simulations produce output on the TB scale. This unprecedented amount of data requires data handling techniques that are beyond what is used for ordinary files. Relational database systems have been successfully used to store and process scientific data, but the new requirements constantly generate new challenges.
Moving terabytes of data among servers on a timely basis is a tough problem, even with the newest high-throughput networks. Thus, moving the computations as close to the data as possible and minimizing the client-server overhead are absolutely necessary.
At least data subsetting and preprocessing have to be done inside the server process. Out of the box commercial database systems perform very well in scientific applications from the prospective of data storage optimization, data retrieval, and memory management but lack basic functionality like handling scientific data structures or enabling advanced math inside the database server. Fortunately, the technology exists to extend the database server with custom-written code that enables us to address these problems.
We present the prototype of a custom-built extension to Microsoft SQL Server that adds array handling functionality to the database system. With our Array Library , fix-sized arrays of all basic numeric data types can be created and manipulated efficiently. With the help of these libraries , complex operations, such as matrix inversions or Fourier transformations, can be done on-the-fly, from SQL code, inside the database server process. We are currently testing the prototype with two different scientific data sets: The Indra cosmological simulation will use it to store particle and density data from N-body simulations, and the Milky Way Laboratory.
This can be used to simulate the neutron capture at incident neutron energies up to 20 MeV to improve the gamma-ray spectrum in neutron data libraries used for transport modelling of unknown assemblies.
Whitefly Bemisia tabaci genome project: Full Text Available Abstract Background The past three decades have witnessed a dramatic increase in interest in the whitefly Bemisia tabaci, owing to its nature as a taxonomically cryptic species, the damage it causes to a large number of herbaceous plants because of its specialized feeding in the phloem, and to its ability to serve as a vector of plant viruses.
Among the most important plant viruses to be transmitted by B. Surprisingly, little is known about the genome of this whitefly. The haploid genome size for male B. The genes involved in whitefly development, in host range plasticity, and in begomovirus vector specificity and competency, are unknown. Results To address this general shortage of genomic sequence information, we have constructed three cDNA libraries from non-viruliferous whiteflies eggs, immature instars, and adults and two from adult insects that fed on tomato plants infected by two geminiviruses: In total, the sequence of 18, clones was determined.
After quality control, and removal of 5, clones of mitochondrial origin 9, sequences remained which included 3, singletons and 1, contigs. Comparisons with public databases indicated that the libraries contained genes involved in cellular and developmental processes. In addition, approximately 1, bases aligned with the genome of the B. Apart from the mitochondrial sequences, the longest and most abundant sequence encodes vitellogenin, which originated from whitefly adult libraries , indicating that much of the gene expression in this insect is directed toward the production.
This study began with the desire to design a public town library of the future and became a search for an inkling of what is essential to Architecture. It is murky and full of contradictions. It asks more than it proposes, and the traces of its windings are better ordered through collage than logical synthesis. This study is neither a thesis nor a synthesis. When drawing out the measure of this study it may be beneficial to state what it attempts to place at the Nuclear data, cross section libraries and their application in nuclear technology.
These proceedings contain the articles presented at the named seminar. The articles deal with evaluated nuclear data libraries , computer codes for neutron transport and reactor calculations using nuclear data libraries , and the application of nuclear data libraries for the calculation of the interaction of neutron beams with materials.
The construction of the multi-group library is realized by using the NJOY nuclear data processing system. This is another in H. Rider Haggard's African series, although this work does not directly involve Alan Quatermain, it nevertheless fills in the history of Umslopogaas of the People of the Ax, who figures materially in the Quatermain works for example "She and Alan". This work is a good read, having much similarity with the other Haggard African novels. There are lots of adventures and several dark villains as well, and the character development brings out that of Galazi, the Wolf, and Mopo, who assumed the role of Umslopogass's father.
We also get a better insight into the characters of Chaka, the father of the Zulu's, and Dingaan. If the reader appreciates 19th century adventure novels, this one is sure to please. HR Haggard's brilliance really shines through with Nada the Lily.
Don't let the name fool you, as Nada really doesn't show up that much. This is much more the story of Umslopogaas, a character from other HRH books. He's a Zulu warrior with an interesting birth scene who, along with his best friend the Wolf, carve a place in literature history.
The action is furious, the deaths are many, and Mopo is a fabulous narrator. I can't recommend this enough - and it's free as it's in the public domain. Grab this if you liked adventure stories. This novel features some characters from some of the Allan Quartermain stories. We have have Umslopogaas, a son of Chaka Zulu, who escaped death and grows up to hate his father. He falls in love with Nada the Lily.
